- 세포 분리부터 iPSC배양까지 18-25일 Protocol

Well Prepared Kit: Reprogramming에 최적화된 제품으로 구성된 종합 키트 

- Step 1: 세포 분리 (RosetteSep™ and SepMate™) 

  Whole blood에서 PBMC (Peripheral Blood Mononuclear Cell)분리 과정에서 mature blood cell 제거를 위한 전용 RosetteSep™ Antibody cocktail이 구성되어 있어 
  분리 단계에서부터 Erythroid progenitor cell의 pre-enrichment 가능

(A) In the donor sample shown above, T-cells (CD3+) represent approximately 21.2% of the PBMC fraction while B-cells (CD19+) are present at 3.4%. 

(B) The addition of the RosetteSep™ Human Progenitor Cell Basic Pre-Enrichment cocktail efficiently depletes the T- and B-cell population to <1% of the enriched cell fraction.

- Step 2: 세포 배양 (StemSpan™ SFEM + Erythroid supplement)

Pre-enrichment Erythroid progenitor cell을 선택적으로 배양을 가능하게하는StemSpan™ SFEM + StemSpan™ Erythroid supplement 가 함께 구성되어 있어

Reprogramming 전 필요한 세포의 수를 늘리는 Post-Expansion이 가능하도록 구성되어 있음

  RosetteSep™ 으로 depletion 한 샘플에서는 CD3+ cell (mature blood cell)의 증가가 거의 없고 CD71+ cell (Erythroid progenitor cell) 의 배양을 최대화 함

- Reprogramming 전용 배지인 ReproTeSR™ 가 함께 구성되어 있어 iPSC colony 형성을 극대화함
