높은 감도로 transcript 검출 및 발현 분석 

2pg, 10pg의 total HEK293 RNA 또는 single cell 샘플을 각각 plexWell Rapid Single Cell RNA-Seq Kit, SMART-Seq v4 Ultra Low input RNA Kit(Takara Bio), Smart-seq2 method로 라이브러리 제작하여 Illumina NextSeq 500에서 2 * 75bp로 시퀀싱 한 결과

극소량 샘플로 high-complexity 라이브러리 제작 가능  

라이브러리는 각각 2pg, 10pg, 100pg, 1ng의 HEK293 total RNA (RIN > 9.0) 샘플을 plexWell Rapid Single RNA-Seq kit 사용하여 제작됨 

극소량의 샘플에서 Transcript 발현과 length 수준에서의 높은 재현성 

라이브러리는 2pg의 total HEK293 RNA (RIN >9.0)으로 각각 plexWell Rapid Single Cell RNA Library prep kit, Takara SMART-Seq v4 Ultra Low Input RNA kit 그리고 Smart-seq2로 제작 됨.

* TPM: Read count로 length를 보정한 정규화된 RNA 발현량 값

PBMCs와 같은 까다로운 샘플에서의 고품질 라이브러리 제작 가능 

BD FACSAria Fusion cell sorter를 이용해 분리된 CD45+ human PBMCs는 plexWell Rapid Kit(녹색, 96 cells), Smart-seq3(빨간색, 96 cells), Smart-seq2(보라색, 48 cells) 그리고 SMART-Seq Single Cell Kit(파란색, 24 cells, Takara)로 라이브러리 제작 됨. 시퀀싱(2 * 75bp)은 Illumina NextSeq에서 진행됨
